چکیده

Dendritic cells (DC) efficiently process and present antigens to the effector arm of the immune system, thereby stimulating immunity against antigens of both foreign and self origin. Administration of Flt3-ligand (FL) has been reported to increase dendritic cell (DC) numbers in mice and humans. As a result, FL has attracted interest as an adjuvant for vaccine immunotherapy. To investigate whether FL might increase the immune response to a model recall antigen, we administered FL 25μg/kg/d subcutaneously to six healthy volunteers followed by a standard injection of intramuscular tetanus toxoid (TT). A control cohort of six healthy volunteers received tetanus toxoid alone. Compared to subjects who received only TT, subjects who received Flt3L and TT Morse#1-vol4no4 12/19/05 10:33 AM Page 536

Improved Immunogenicity of Tetanus Toxoid by Brucella abortus S19 LPS Adjuvant

Background: Adjuvants are used to increase the immunogenicity of new generation vaccines, especially those based on recombinant proteins. Despite immunostimulatory properties, the use of bacterial lipopolysaccharide (LPS) as an adjuvant has been hampered due to its toxicity and pyrogenicity. Brucella abortus LPS is less toxic and has no pyrogenic properties compared to LPS from other gram nega...

FLT3 ligand enhances the cancer therapeutic potency of naked RNA vaccines.

Intranodal immunization with antigen-encoding naked RNA may offer a simple and safe approach to induce antitumor immunity. RNA taken up by nodal dendritic cells (DC) coactivates toll-like receptor (TLR) signaling that will prime and expand antigen-specific T cells. In this study, we show that RNA vaccination can be optimized by coadministration of the DC-activating Fms-like tyrosine kinase 3 (F...

Mucosal Adjuvant Potential of Quillaja saponins and Cross-linked Dextran Microspheres, Co-administered with Liposomes Encapsulated with Tetanus Toxoid

Intranasal vaccination is particularly a striking route for mucosal immunization, due to the ease of administration and the induction of both mucosal and humoral immunity. However, soluble antigens (Ag) are not sufficiently taken up after the nasal administration and need to be co-administered with adjuvants, penetration enhancers or encapsulated in particles. So, in this study, tetanus toxoid ...

Nasal Immunization by (PLGA) Nanospheres Encapsulated with Tetanus Toxoid and (CpG-ODN)

In induction of systemic and mucosal immunity, particulate antigens are more effective than soluble antigens possibly because they are more efficiently endocytosed by mucosal-associated lymphoid tissue (MALT) M cells. In this study, we determined the systemic and mucosal immune responses in rabbits following intranasal immunization of tetanus toxoid TT and CpG-ODN encapsulated within PLGA nanos...
